چکیده

In this research, post-thaw effects of bull semen extenders including different amounts of boron (sodium pentaborate) on total motility (TMOT), progressive motility (PMOT), kinetic parameters of spermatozoa (VCL, VSL, VAP, LIN, STR, WOB, ALH, BCF), plasma membrane and acrosome integrity (PMAI) and DNA fragmentation index (DFI) were evaluated. Eight ejaculates were examined from four different bulls. Each ejaculate was split in to five aliquots and extended with standard Tris extender (Control) and 0.4 g boron with 0.5 g glucose (Group 1) 0.5; 0.7; 0.9 g boron without glucose (Group 2, Group 3 and Group 4 respectively) were used as experimental groups. After thawing TMOT, PMOT, kinetic traits of spermatozoa, PMAI and DFI were determined. According to motility and progressive motility (66.04%, 50.79%), Group 1 had higher values than the others, but differences between groups were not statistically significant (P>0.05). The highest values of VSL, LIN, STR and WOB were determined (28.15 μm/s, 42.98%, 71.69%, 59.71%) in Group 1 (P<0.05). While other kinetic traits of spermatozoa were higher numerically in Group 1 than the other groups, differences between groups were not statistically significant (P>0.05). The highest PMAI value was determined as 60.84% in Group 1 (P>0.05). The DFI rates were found similar in all control and experimental groups (P>0.05). In conclusion, it was observed that the boron (sodium pentaborate) has positive effects on post-thaw movement and structural traits. In addition, boron has no detrimental effect on DNA fragmentation index in semen freezing. Thus, boron could be used as an alternative semen extender compound.

Effect of the Addition of Insulin Like Growth Factor-I (IGF-I) on Cryopreservation of Hariana Bull Semen

This experiment was designed to study the effect of insulin like growth factor-I (IGF-I) on cryopreservation of Hariana bull semen. For this purpose, semen ejaculates were collected from four Hariana bulls using artificial vagina at biweekly interval. The semen samples which possess more than 70% initial progressive motility and above 500 million/mL spermatozoa concentration was subsequently su...

O-12: Studies on Sequestration of PDC-109 Protein on Cryodamage and In Vitro Fertility of Crossbred Bull Spermatozoa

Background: Plasma membrane of the spermatozoa interacts with and is altered by abundant seminal PDC-109 present in their immediate milieu in vivo with damaging effect in a time- and concentration- dependent manner. Therefore we hypothesized that sequestration of detrimental protein from ejaculates would be beneficial for cryopreservation of sperm cells. To this aim we evaluated the effect of s...

Effect of pre-freeze semen quality, extender and cryoprotectant on the post-thaw quality of Asian elephant (Elephas maximus indicus) semen.

Semen cryopreservation and artificial insemination (AI) are potentially valuable methods for supporting the breeding management of endangered species like the Asian elephant. Cryopreservation of Asian elephant semen has however proven problematic with respect to maintenance of both adequate semen quality and fertility post-thaw. In this study, nine ejaculates from three adult bulls were used to...

Influence of Bull Seminal Plasma on Post-thaw Ram Semen Parameters and Fertility

The aim of the study was to evaluate the effect of the addition of 20% bull seminal plasma to semen extenders on post-thaw sperm motility, morphology, and fertility. Two Merino rams at 3-5 years of age, 43 ewes, and one Holstein bull were used. Fourteen ejaculates were collected by the use of an artificial vagina, twice a day, at 60 min interval. Six semen ejaculates were extended with Tris-egg...

Effect of Non-enzymatic Antioxidants in Extender on Post-thaw Quality of Buffalo (bubalus Bubalis) Bull Spermatozoa

The objective of the present study was to determine the effect of non-enzymatic antioxidants (vitamin C or E) in tris-citric acid buffer (TCA) on post-thaw quality of buffalo (Bubalus bubalis) bull spermatozoa. Split pooled buffalo bull ejaculates were diluted in TCA egg yolk glycerol extender containing either vitamin C (TCAC), vitamin E (TCAE) or without antioxidant (TCAN) at 37°C. Extended s...
